I use Avast, and it has failed me, actually. The virus is still there. I've put a total of two whole days into it.
At this point, I'm going to turn Avast off (because I'm tired of getting the "Virus detected!" message every five minutes), and allow my computer to destroy itself from the inside-out until this weekend, when I can reinstall windows.
Wait! You should always try more than one anti-virus, do not ever -- I repeat, ever -- rely on just one anti-virus. Otherwise, you're going to do exactly what you're about to do and lose everything. Try cross-referencing with one of the other programs mentioned before you do anything drastic. Also, don't forget that not every anti-virus is perfect (also another reason to cross-reference) and it could be just as much a false alarm as it is a real issue. So you should definitely cross reference before you defrag/restore/etc, Nate.
